CJC-1295 without DAC (Mod GRF 1-29)
Tetrasubstituted GHRH(1-29) amide without DAC (Mod GRF 1-29) — investigational; not FDA-approved.
Editorial review:Evidence snapshot
This drawer marks papers. It is not a plan, a dose, or a source. A paper is a bounded report. It is not a protocol. Grades name the object. They are not a safety rating.
Identity lock — SEPARATE from with-DAC. This object is CJC-1295 without DAC, also called Mod GRF 1-29: a tetrasubstituted GHRH(1-29) amide (commonly described substitutions include D-Ala², Gln⁸, Ala¹⁵, Leu²⁷) without the Drug Affinity Complex / maleimidopropionyl-Lys albumin-binding linker. It is not the long-acting albumin-binding analog studied as CJC-1295 in Teichman / Ionescu / Sackmann-Sala. It is not native sermorelin / GHRH(1-29). PubChem CID 56841945 (formula C152H252N44O42, MW ~3367.9) carries synonym noise that also lists “with DAC” language — do not trust a single CAS badge. CAS strings 863288-34-0, 446262-90-4, and 446036-97-1 collide across with-DAC / without-DAC synonym lists → To be confirmed; do not invent one CAS. No approved indication. Off-label does not apply. There is no label to be off of.
Bottom line
Verified human PK/PD or efficacy trials that cleanly name without-DAC / Mod GRF 1-29 as the intervention object are essentially absent from this PubMed set. Do not borrow Teichman 2006, Ionescu 2006, or Sackmann-Sala 2009 — those describe the long-acting albumin-binding (with DAC) analog. Detection and narrative papers that mention “CJC-1295” without DAC status are naming noise, not without-DAC efficacy. Honest grade: GH-axis pharmacology is plausible by class (GHRH analog), but human evidence for this specific object is not established here.
Papers
Identity contrast — sibling object (with DAC), not a without-DAC grade
PMID 15817669 — Jetté 2005
Tested: maleimido hGRF(1-29)–albumin bioconjugates in rats; identifies CJC-1295 as tetrasubstituted hGRF(1-29) with an Nε-3-maleimidopropionamide lysine (albumin-binding / DAC-class linker) and extended plasma presence.
Does not show: human without-DAC / Mod GRF 1-29 PK/PD. Use only to lock that with-DAC ≠ without-DAC.
What this set does not contain (load-bearing)
No verified ascending-dose healthy-volunteer PK/PD, growth, body-composition, or disease RCT in this set that cleanly labels CJC-1295 without DAC / Mod GRF 1-29 as the administered peptide. Papers graded under the with-DAC drawer (Teichman 16352683, Ionescu 17018654, Sackmann-Sala 19386527, Alba 16822960) must not be re-badged here.

Tested: antibody-free ultrafiltration assay for GHRH analogs in urine.
Does not show: a clinical endpoint for Mod GRF 1-29.
PMID 42395176 — Dominikowski 2026
Tested: narrative review of performance-enhancing peptides on the GH–IGF-I axis; clinical evidence described as limited relative to self-administration culture.
Does not show: a new controlled without-DAC trial.
What is not established
Any approved use. Human PK/PD equivalence to CJC-1295 with DAC. Body composition, recovery, sleep, or “anti-aging” as treatment claims for without-DAC. A single printable CAS (PubChem synonym collision). Product identity between any vial labeled “CJC-1295” and either DAC status. Do not merge paper grades across the two drawers.
