CJC-1295 with DAC
Long-acting albumin-binding GHRH(1-29) analog with DAC — investigational; not FDA-approved.
Editorial review:Evidence snapshot
This drawer marks papers. It is not a plan, a dose, or a source. A paper is a bounded report. It is not a protocol. Grades name the object. They are not a safety rating.
Identity lock — SEPARATE from without-DAC / Mod GRF 1-29. This object is CJC-1295 with DAC (Drug Affinity Complex): a tetrasubstituted GHRH(1-29) analog carrying a maleimidopropionyl-Lys linker that covalently binds serum albumin, yielding a long-acting GHRH receptor agonist. PubChem CID 91971820 (also 172966534); formula C165H269N47O46; MW ~3647.2. CAS strings often cited (863288-34-0, 446262-90-4, and others) also appear on without-DAC synonym lists → print CAS as To be confirmed; do not invent one locked CAS for UI. Not sermorelin. Not Mod GRF 1-29 without the DAC linker. No approved indication. Off-label does not apply. There is no label to be off of.
Bottom line
Thin but real human PK/PD exists for the albumin-binding long-acting analog: healthy-adult trials show prolonged GH and IGF-I rises with preserved GH pulsatility. That is a GH-axis stimulation object in volunteers — not an approved indication, not body composition, not anti-aging, and not evidence for without-DAC / Mod GRF 1-29. Preclinical growth rescue appears in a GHRH-knockout mouse paper. Detection/doping papers are not efficacy.
Papers
Identity / discovery — albumin-binding analog
PMID 15817669 — Jetté 2005
Tested: maleimido hGRF(1-29)–albumin bioconjugates; identifies CJC-1295 as tetrasubstituted hGRF(1-29) with Nε-3-maleimidopropionamide lysine; rat GH AUC and multi-day plasma presence.
Does not show: a human therapeutic indication, or results for the without-DAC peptide alone.
Early human — with DAC
PMID 16352683 — Teichman 2006
Tested: CJC-1295 (long-acting GHRH analog that binds albumin; reported multi-day half-life) versus placebo in healthy adults — ascending single-dose and multi-dose PK/PD for GH and IGF-I.
Does not show: an approved therapeutic use, a body-composition claim, or without-DAC / Mod GRF 1-29 results.
PMID 17018654 — Ionescu 2006
Tested: overnight GH pulsatility in healthy young men before and after a single injection of the same long-acting CJC-1295 analog; trough/mean GH and IGF-I.
Does not show: a disease-treatment trial, or that without-DAC Mod GRF produces the same multi-day profile.
PMID 19386527 — Sackmann-Sala 2009
Tested: serum proteomic changes in healthy young men one week after CJC-1295 (long-acting GHRH analog) injection; exploratory GH/IGF-I action biomarkers.
Does not show: clinical benefit, an approved use, or a without-DAC result. Small n.
Preclinical — with DAC
PMID 16822960 — Alba 2006
Tested: once-daily CJC-1295 (long-acting GHRH analog) in GHRH knockout mice for growth normalization.
Does not show: a human growth or adult-body-composition result.
Context, not a grade — detection / narrative
PMID 21871962 — Thomas 2012
Tested: immunoaffinity purification of peptide hormones before LC-MS in doping controls.
Does not show: human therapeutic efficacy.
PMID 26879649 — Knoop 2016
Tested: qualitative identification of GHRH analogs in human plasma.
Does not show: clinical benefit.
PMID 34665524 — Memdouh 2021
Tested: advances in detection of synthetic GHRH analogs.
Does not show: efficacy.
PMID 35298973 — Coppieters 2022
Tested: antibody-free ultrafiltration assay for GHRH analogs in urine.
Does not show: a clinical endpoint.
PMID 42395176 — Dominikowski 2026
Tested: narrative review of performance-enhancing peptides modulating the GH–IGF-I axis; clinical evidence limited relative to marketing/self-administration culture.
Does not show: a new controlled human CJC-1295 with-DAC efficacy trial for physique or anti-aging claims.
What is not established
Any approved indication. Body fat, muscle, recovery, or “anti-aging” as human treatment claims. Equivalence of with-DAC and without-DAC products. A single collision-free CAS for UI. Identity between trial material and any circulating vial labeled only “CJC-1295.”
