Investigational / not FDA-approved

GHRP-6

Classic Bowers GH-releasing hexapeptide — investigational; not FDA-approved.

Editorial review:
What evidence says

Evidence snapshot

This drawer marks papers. It is not a plan, a dose, or a source. A paper is a bounded report. It is not a protocol. Grades name the object. They are not a safety rating.

Identity lock. GHRP-6 is the classic synthetic GH-releasing hexapeptide His-D-Trp-Ala-Trp-D-Phe-Lys-NH₂. PubChem CID 4345065; formula C46H56N12O6; MW ~873.0; CAS 87616-84-0. PubChem synonym noise may list unrelated strings (including “GHRP-2 Acetate”-style errors) — trust sequence/CAS/CID, not synonym spam. It is not hexarelin (2-methyl-Trp analog), not GHRP-2/pralmorelin, and not ipamorelin. Foundational Bowers-line discovery and human synergy-with-GHRH work define this object. No approved bodybuilding indication. Off-label does not apply. There is no label to be off of.

Bottom line

Strong foundational human and preclinical GH-secretagogue literature: GHRP-6 releases GH, acts synergistically with GHRH in many settings, and entered adult GHD diagnostic algorithms (alone or with GHRH) as an alternative stimulus to insulin tolerance testing in research/clinical endocrinology. Later organ-failure and animal models are not physique grades. Anti-doping detection papers are not efficacy. Keep hexarelin / GHRP-2 / ipamorelin out of this drawer’s paper grades.

Papers

Discovery / early human — Bowers line

PMID 6714155 — Bowers 1984

Tested: in vitro and in vivo activity of a synthetic hexapeptide that specifically releases GH from the pituitary (foundational GHRP object matching the GHRP-6 sequence family).

Does not show: modern outpatient anti-aging efficacy, or identity with GHRP-2/hexarelin.

PMID 2108187 — Bowers 1990

Tested: GHRP stimulates GH release in normal men and acts synergistically with GHRH.

Does not show: a chronic therapy indication or body-composition RCT.

PMID 1730807 — Bowers 1992

Tested: oral administration of the GH-releasing hexapeptide (His-DTrp-Ala-Trp-DPhe-Lys-NH₂) in normal men and short-statured children; GH and peptide levels.

Does not show: approved oral secretagogue product for wellness use, or hexarelin results.

PMID 8370708 — Alster 1993

Tested: GH response to GHRP (His-DTrp-Ala-Trp-DPhe-Lys-NH₂), GHRH, and TRH in men (specificity / stimulus characterization).

Does not show: chronic treatment benefit.

Human — stimulus physiology / aging / pathology

PMID 8421084 — Peñalva 1993

Tested: effects of GHRH, atropine, pyridostigmine, or hypoglycemia on GHRP-6-induced GH secretion in man (mechanistic endocrine physiology).

Does not show: a therapeutic body-composition claim.

PMID 7734029 — Micic 1995

Tested: GH secretion after GHRP-6 or GHRH+GHRP-6; whether responses decline in late adulthood.

Does not show: anti-aging treatment efficacy.

PMID 7883854 — Popovic 1995

Tested: blocked GHRP-6-induced GH secretion and loss of GHRP-6+GHRH synergy in hypothalamic-pituitary disconnection / related pathology contexts.

Does not show: recreational use guidance; diagnostic/pathophysiology object.

Human — diagnostic GHD testing

PMID 8887178 — Pombo 1996

Tested: GHRP-6 test in the diagnosis of GH deficiency.

Does not show: chronic replacement with GHRP-6 as standard of care, or physique outcomes.

PMID 11980622 — Petersenn 2002

Tested: diagnosis of adult GH deficiency with GHRP-6 alone or combined with GHRH versus insulin tolerance test.

Does not show: approved long-term secretagogue therapy for non-GHD adults.

PMID 15817913 — Haijma 2005

Tested: GHRH/GHRP-6 test for GHD diagnosis in elderly or severely obese men.

Does not show: obesity treatment with GHRP-6.

Context — organ-failure model / detection

PMID 16417467 — Cibrián 2006

Tested: GHRP-6 for prevention of multiple organ failure (translational/clinical exploratory object — not a gym indication).

Does not show: bodybuilding efficacy or an approved ICU labeling grade in this pack’s framing.

PMID 25809000 — Cox 2015

Tested: detection of GHRP-2 and GHRP-6 in athlete urine.

Does not show: therapeutic benefit.

What is not established

Approved anti-aging, muscle-building, or fat-loss indications. Equivalence to hexarelin, GHRP-2, or ipamorelin. Chronic unsupervised outpatient use as “therapy.” Product identity for unlabeled vials. Diagnostic GH release is not a physique protocol and is not graded as one.
