Mitochondria-derived 24-aa peptide (MT-RNR2) — ≠ MOTS-c ≠ HNG; no human outcome RCTs here.
Editorial review:Evidence snapshot
This drawer marks papers. It is not a plan, a dose, or a source. A paper is a bounded report. It is not a protocol. Grades name the object. They are not a safety rating.
Identity lock. Humanin is a mitochondria-derived 24-amino-acid peptide encoded in the mitochondrial MT-RNR2 region (UniProt Q8IVG9; PubChem CID 16131438; formula C119H204N34O32S2 / MW 2687.2; CAS 330936-69-1; sequence often given as MAPRGFSCLLLLTSEIDLPVKRRA). Related analogs (e.g. HNG / S14G-humanin, colivelin) are different objects. ≠ MOTS-c (separate mitochondria-derived peptide drawer elsewhere). No approved human therapeutic indication in this pack’s framing. This drawer marks papers. It is not use advice.
Bottom line
Discovery-era neuroprotection papers (Hashimoto PNAS/J Neurosci 2001) established humanin as a secreted rescue factor against Alzheimer-relevant insults in cell/animal models. Receptor and IGFBP-3 interaction biology followed. Translation reviews discuss cardiometabolic and aging hypotheses. Controlled human disease-outcome trials for humanin peptide administration are not established in this Selected sources set — refuse anti-aging glow.

Does not show: a human clinical Alzheimer therapy.
PMID 11717357 — Hashimoto 2001
Tested: detailed characterization of neuroprotection by humanin against Alzheimer-relevant insults.
Does not show: human RCT efficacy.
PMID 11327724 — Hashimoto 2001
Tested: mechanisms of neuroprotection by humanin from Swedish mutant APP models.
Does not show: approved neuroprotective drug status.

Tested: interaction between humanin and IGFBP-3 regulating cell survival/apoptosis.
Does not show: human metabolic therapy outcomes.
Receptors / class reviews
PMID 19997871 — Matsuoka 2010
Tested: review of humanin and proposed receptors for humanin.
Does not show: a completed pivotal clinical package.
PMID 16958985 — Matsuoka 2006
Tested: review of humanin and colivelin as neuronal-death-suppressing peptides (Alzheimer/ALS framing) — colivelin is a related but different object.
Does not show: human approval for either peptide.
PMID 27082450 — Xiao 2016
Tested: review of humanin functional interfaces with IGF-I.
Does not show: IGF-1 LR3 or IGF DES equivalence.
PMID 32800320 — Rochette 2020
Tested: emerging-properties review of humanin as a mitochondria-derived peptide (cardiology-adjacent narrative).
Does not show: cardiovascular outcome trial proof.
Exploratory / variant context
PMID 38757793 — Logan 2024
Tested: humanin P3S / haplogroup N1b genetic epidemiology framing and Alzheimer risk — endogenous variant epidemiology, not administered-peptide efficacy.
Does not show: that supplemental humanin treats Alzheimer disease.
PMID 39079574 — Wang 2024
Tested: preclinical reproductive-damage model examining humanin and MOTS-c — MOTS-c is a different MDP object.
Does not show: human fertility therapy for humanin.
What is not established
Human clinical disease-modifying efficacy for administered humanin. Interchangeability with HNG, colivelin, or MOTS-c. Anti-aging or body-composition claims from forum use. Product identity for unregulated humanin preparations. This drawer does not provide use advice, schedules, or sourcing.
