Melatonin
Endogenous indoleamine chronobiotic — ≠ Melanotan; Circadin PR ≠ all OTC melatonin pathways.
Editorial review:Evidence snapshot
This drawer marks papers. It is not a plan, a dose, or a source. A paper is a bounded report. It is not a protocol. Grades name the object. They are not a safety rating.
Identity lock. Melatonin (N-acetyl-5-methoxytryptamine) is an endogenous pineal indoleamine chronobiotic. PubChem CID 896; formula C13H16N2O2 / MW 232.28; CAS 73-31-4. ≠ Melanotan (α-MSH analogue naming collision is accidental English overlap only). Pharmaceutical prolonged-release melatonin (Circadin-class) is a regulated insomnia object in some jurisdictions; dietary-supplement melatonin is a different regulatory pathway in others. This drawer marks papers. It is not use advice.
Bottom line
Sleep meta-analyses support modest effects on selected sleep outcomes for melatonin, with heterogeneity by population, formulation, and timing. Prolonged-release melatonin has labeled/specialty insomnia framing (including Circadin-class reviews and ASD pediatric prolonged-release programs). Chronobiology reviews explain mechanism without inventing unlimited wellness claims. Melatonin is not Melanotan.

Tested: meta-analysis of melatonin for primary sleep disorders.
Does not show: that every OTC product matches trial formulations, or Melanotan identity.
PMID 33417003 — Fatemeh 2022
Tested: systematic review and meta-analysis of melatonin supplementation on sleep quality (RCT pool).
Does not show: unlimited jet-lag or shift-work proof for every regimen studied elsewhere.

PMID 38888087 — Cruz-Sanabria 2024
Tested: systematic review and dose-response meta-analysis optimizing time/dose framing for melatonin as a sleep-promoting drug — methods paper on published trials; this drawer still provides no use schedule.
Does not show: a universal consumer protocol endorsement.
PMID 37434463 — Maruani 2023
Tested: systematic review/meta-analysis of melatonin and ramelteon for acute and long-term insomnia disorder management (ramelteon is a different MT1/MT2 receptor agonist drug).
Does not show: that melatonin equals ramelteon.

Tested: review of melatonin in human sleep, circadian rhythms, and regulation.
Does not show: new primary RCT data by itself.
PMID 25908646 — Claustrat 2015
Tested: review of physiological effects of melatonin in humans.
Does not show: cosmetic or athletic claims.

Tested: review of melatonin as a chronobiotic with sleep-promoting properties.
Does not show: approval status in every jurisdiction.
PMID 39400423 — Comai 2024
Tested: review moving beyond traditional views of melatonin, receptors, and sleep.
Does not show: a new pivotal insomnia registration trial.
Prolonged-release / labeled-adjacent pharmaceutical framing
PMID 22429105 — Lemoine 2012
Tested: review of prolonged-release melatonin (Circadin) for insomnia treatment.
Does not show: that immediate-release dietary supplements equal the prolonged-release pharmaceutical object.
PMID 29096777 — Gringras 2017
Tested: efficacy and safety RCT of pediatric prolonged-release melatonin for insomnia in children with autism spectrum disorder.
Does not show: adult OTC equivalence or Melanotan-related biology.
What is not established
That all melatonin products are interchangeable with Circadin-class prolonged-release. That meta-analytic sleep benefits equal treatment of every circadian disorder. Any connection to Melanotan I/II chemistry. Unlimited anti-aging or immune claims. This drawer does not provide use advice, schedules, or sourcing.
